Yeast System
Yeast is a single-celled eukaryotic organism capable of producing very large quantities of recombinant protein, it offer many of the advantages of producing proteins in microbes (growth speed, easy genetic manipulation, low cost media) while offering some of the attributes of higher eukaryotic systems (post translational modifications, secretory expression). Several yeast protein expression systems exist in organisms from the genera Saccharomyces, Pichia, Kluyveromyces, Hansenula and Yarrowia. Our two-phase system for recombinant protein expression includes subcloning into the expression vector, generating and identifying the expression of positive clones.

Description:

  • Amplification/isolation of the gene of interest from a customer-supplied vector and subcloning into a suitable expression vector

  • 2 weeks; If insert > 2kb, an additional week per 1kb

Phase II: Generation and Identification of Expression Positive Clones

Description:

  • Transformation of recombinant constructs into a suitable yeast strain
  • Small scale induction to over-express target protein
  • Test for recombinant protein by SDS-PAGE and western blot (customer must provide appropriate antibodies)
